RADIOLIGAND BINDING ANALYSIS

Kell screenshotKELL is one of the most-cited computer packages in the scientific literature and forms the basis of radioligand binding studies in thousands of laboratories around the world. The package was first described in J. Pharmacol. Methods 14, 213-228, 1985, and has been regularly upgraded since then.

KELL uses weighted non-linear, iterative curve-fitting techniques to analyze saturation and competition equilibrium studies and it can also determine association and dissociation rate constants. Parameter estimates can be fixed or limited. Appropriateness of models is determined by built-in statistics tests which include F-test and runs test. A wide range of graphs includes plots of the saturation data as the binding isotherm as well as Scatchard and Hill plots.

KELL also incorporates a user-friendly version of the well-known LIGAND program devised by Munson and Rodbard, which remains the tool of choice for sophisticated curve-fitting involving multiple ligands binding to multiple sites.

Data entry is straightforward and can be made directly via the keyboard or by pasting data from spreadsheets. Any number of datapoints and replicates can be used and the number of ligands and receptors which can be simultaneously handled is limited only by hardware.

Another feature is the built-in general assay analyzer which incorporates a range of standard curve models including linear, hyperbolic, incremental exponential and decremental exponential. In the context of ligand binding studies it can be used to calculate protein concentrations, but it is useful for analyzing almost any assay which uses standard curves.

All numeric and graphical results can easily be transferred to other programs. The fitted data of KELL graphs can optionally be exported for replotting in your usual graphics package. There is a manual with tutorials and also built-in Help.
